C4: Porcine islet macroencapsulation – a concept for xenogeneic islet transplantation
The proof-of-concept of islet macroencapsulation using the Beta-O2 device has been demonstrated in large animals and in a first patient study, and provided sufficient immune isolation whilst allowing regulated islet graft function to be maintained. In a first application of porcine islet xenografts in diabetic NHP, we demonstrated a favourable safety profile and persistent graft function without any immunosuppression. Based upon this, a first clinical trial of macroencapsulated porcine islets in man is being targeted for the second funding period. In addition, several structural aspects of the current device will be optimized for larger scale application and alternative cell sources with a particular focus on evaluating isolation efficacy, functional potency and immunogenicity.
